How reconstitution maths works
- Divide the vial amount by the water you added to get the concentration, then divide your dose by that concentration to get the volume. A 5 mg vial in 2 ml of bacteriostatic water is 2.5 mg/ml, or 2500 mcg/ml. A 250 mcg dose is therefore 0.1 ml, which is 10 units on a U-100 insulin syringe.
- No — and this is the single most common misunderstanding. The vial contains a fixed amount of peptide however much water you add. More water simply means the same dose occupies a larger, easier-to-measure volume. It changes the draw, not the amount of compound.
- On a standard U-100 insulin syringe, 100 units equals 1 ml, so one unit is 0.01 ml. The units are volume markings, not a measure of how much peptide you are getting — that depends entirely on your concentration.
- Enough that your intended dose lands on an easily readable mark, ideally 10 units or more. Very small draws of one or two units are difficult to measure accurately, and measurement error is proportionally large. The calculator suggests a volume that puts your dose on the 10-unit mark.
